When you use the matchmaking service Unity will try to directly connect to the server. When using Unity's matchmaking service (which is a paid service) all clients and clients who start a local server will connect to the matchmaking server in order to find a match they can join. Setting up a valid route through the internet is independent from creating a network application. However if you want to connect through the internet the server has to port forward the port that your application is using and the client has to use the public IP address of the server (the IP address of the router). On a local area network this is pretty simple. You just have to specify an IP address "you can reach" and connect to that. Connection between different devices (especially through the internet) is nothing Unity specific. However running one client (or the server) in the editor makes it easier to debug the project. Generally it doesn't matter if he uses the editor and one build version of the game or two build versions of the game.
